چکیده

(-)-Epigallocatechin-3-gallate (EGCG) has exhibited been studied for lung cancer inhibitory activity in vitro and in animal models, but it is rapidly methylated and inactivated by catechol-O-methyltransferase (COMT). Entacapone and tolcapone, COMT inhibitors, are used to mitigate the symptoms of Parkinson's disease. We investigated the synergistic effects of entacapone/tolcapone and EGCG against lung cancer cell lines in culture. EGCG, entacapone and tolcapone inhibited the growth of H1299 human lung cancer cells (IC50 = 174.9, 76.8 and 29.3 µM, respectively) and CL-13 murine lung cancer cells (IC50 = 181.5, 50.7 and 19.7 µM, respectively) as single agents following treatment for 72h. Treatment with 1:10, 1:5, 1:2.5 and 1:1 combinations of EGCG and tolcapone or entacapone resulted in synergistically enhanced growth inhibition. The growth inhibitory effect of the combinations was mediated by induction of intracellular oxidative stress, cell cycle arrest and decreased nuclear translocation of nuclear factor-κΒ. Methylation of EGCG was dose dependently inhibited by entacapone and tolcapone (IC50 = 10 and 20 µM, respectively) in a cell-free system, and both compounds increased the intracellular levels of unmethylated EGCG. Treatment of mice with EGCG in combination with tolcapone increased the bioavailability of EGCG and decreased the methylation of plasma norepinephrine: no apparent liver or behavioral toxicity was observed. In conclusion, the combination of EGCG and entacapone/tolcapone synergistically inhibited the growth of lung cancer cells in culture, and the mechanistic basis for this synergy is likely due in part to inhibition of COMT with resultant increase in the levels of unmetabolized EGCG.

Enzymology of methylation of tea catechins and inhibition of catechol-O-methyltransferase by (-)-epigallocatechin gallate.

(-)-Epigallocatechin gallate (EGCG) and (-)-epigallocatechin (EGC) are the major polyphenolic constituents in green tea. In this study, we characterized the enzymology of cytosolic catechol-O-methyltransferase (COMT)-catalyzed methylation of EGCG and EGC in humans, mice, and rats. At 1 microM, EGCG was readily methylated by liver cytosolic COMT to 4"-O-methyl-EGCG and then to 4',4"-di-O-methyl-...

Quercetin increased bioavailability and decreased methylation of green tea polyphenols in vitro and in vivo.

The extensive methylation of green tea polyphenols (GTPs) in vivo may limit their chemopreventive potential. We investigated whether quercetin, a natural inhibitor of catechol-O-methyltransferase (COMT) and multidrug resistance proteins (MRPs), will differentially increase the intracellular concentration and decrease the methylation of GTPs in different cancer cell lines. Intrinsic COMT activit...

Green tea extract thermogenesis-induced weight loss by epigallocatechin gallate inhibition of catechol-O-methyltransferase.

Epidemiological studies have shown that intake of tea catechins is associated with a lower risk of cardiovascular disease. The antioxidative activity of tea-derived catechins has been extensively studied. Reports have shown that green tea extract intake is associated with increased weight loss due to diet-induced thermogenesis, which is generally attributed to the catechin epigallocatechin gall...

O-Methylation of tea polyphenols catalyzed by human placental cytosolic catechol-O-methyltransferase.

In the present study, we evaluated the metabolic O-methylation of several catechol-containing tea polyphenols by human placental catechol-O-methyltransferase (COMT). (-)-Epicatechin, (+)-epicatechin, and (-)-epigallocatechin were good substrates for metabolic O-methylation by placental cytosolic COMT (150-500 pmol/mg of protein/min), but (-)-epicatechin gallate and (-)-epigallocatechin gallate ...

Reversal of hypermethylation and reactivation of genes by dietary polyphenolic compounds.

Dietary polyphenolic compounds have been reported to have many interesting biological activities, including the induction of epigenetic changes and cancer prevention. In searching for the mechanisms of the anti-cancer action of (–)–epigallocatechin-3-gallate (EGCG), the major polyphenolic compound in green tea, we observed the inhibition of DNAmethyltransferase (DNMT) activity by EGCG.1 Treatme...
